    Scalable Observation, Analysis, and Tuning for Parallel Portability in HPC

    It is desirable for general productivity that high-performance computing applications be portable to new architectures, or can be optimized for new workflows and input types, without the need for costly code interventions or algorithmic re-writes. Parallel portability programming models provide the potential for high performance and productivity, however they come with a multitude of runtime parameters that can have significant impact on execution performance. Selecting the optimal set of parameters, so that HPC applications perform well in different system environments and on different input data sets, is not trivial.This dissertation maps out a vision for addressing this parallel portability challenge, and then demonstrates this plan through an effective combination of observability, analysis, and in situ machine learning techniques. A platform for general-purpose observation in HPC contexts is investigated, along with support for its use in human-in-the-loop performance understanding and analysis. The dissertation culminates in a demonstration of lessons learned in order to provide automated tuning of HPC applications utilizing parallel portability frameworks

    Fluency Assistance Device (FAD): Masker Upgrades

    There are around seventy million people internationally who have a stutter, a form of fluency disorder. Some fluency assistance devices are available to the public, but most are highly expensive or unreliable. The Fluency Assistive Device (FAD) team seeks to assist a niche community of these individuals for whom therapy has not worked, and who currently rely on a device known as the Edinburgh Masker. To best reach this community, FAD is partnering with Dave Germeyer, who has invaluable experience repairing these masker devices for his clientele. FAD is redesigning the masker to increase its portability, functionality, and cost-effectiveness by developing an improved analog and new digital version. The Analog Masker v1.3 focuses on updated components and consolidated circuitry to eliminate troublesome wiring of the original. The Digital Masker v1.0 employs a Bluetooth-enabled microcontroller to achieve masker functionality, offering the flexibility of alternative fluency assistance algorithms to assist a broader group of users. An updated prototype of the Analog Masker v1.3 was fabricated and tested for power consumption and overall functional output characteristics versus the original Edinburgh version. The Analog Masker v1.3 has also been fully packaged and enclosed to produce a client testable unit. Bluetooth audio output for the Digital Masker has almost been completed, and two of the alternative algorithms have been coded for the masking output. One of these algorithms, Delayed Altered Feedback (DAF), now produces the expected output in response to an audio test input. Clarity and integrity of the DAF signal output have also been improved. The Masking Altered Feedback (MAF) algorithm that emulates the behavior of the Edinburgh original on the Digital Masker v1.0 is under development.     Development of the Continuously Variable Volume Reactor for Flow Injection AnalysisDesign, Capabilities and Testing

    A new apparatus for mixing sample and reagent in flow injection analysis is described. The continuously variable volume reactor (CVVR) replaces the conventional mixing coil in a flow injection manifold to provide mixing and dilution. A linear actuator motor allows control of the chamber volume via Lab VIEW software. The chamber volume can be incremented in steps of 1 uL over the range 68-1704 uL. In addition, the chamber has an integral variable-speed stirring unit that is also under computer control. Experiments were performed to evaluate the dispersion characteristics of this new device, evaluate the volume reproducibility, and understand the mixing characteristics. Use of the chamber is shown in the determination of iron (II) in pond water, and in NIST SRM 1643d with excellent results and a detection limit of 3.7 ug/L iron(II). Advantages of the CVVR and future research activities using the device are discussed

    Development of a Geothermal Well Database for Estimating In-Field EGS Potential in the State of Nevada

    ABSTRACT Recent successes at Department of Energy (DOE)-funded Enhanced Geothermal System (EGS) demonstration projects have shown that EGS techniques can be used in existing hydrothermal fields to enhance the permeability of geothermal wells, converting previously "dry" wells into commercially-viable ones (Desert Peak EGS Project, DOE Award: DE-FC6-02ID14406). The application of EGS techniques to dry wells in existing hydrothermal fields, defined here as "in-field EGS," has the potential to increase or help maintain generation capacity at hydrothermal power plants. The amount of in-field EGS potential depends on the number and status of dry wells at existing hydrothermal power plants, and their location with respect to the geothermal reservoir and other geothermal wells in the field. To aid in estimating the in-field EGS potential, the authors are developing a database containing and consolidating the publically available information on full-sized geothermal wells at hydrothermal power plants. The goal of this database development was to identify the name, location, and status of all full-sized geothermal wells drilled to date. Early design and population of the database focused on wells at hydrothermal power plants in Nevada. The database was created by aggregating and cleaning data from publically available datasets. The initial database was then examined for missing or possibly erroneous data and corrected using original source documents such as well logs, permitting documents, etc. and the data source of the information updated as well. The database was designed to track data sources for each well data point, so that information can be traced back to its original source. The resulting design allows the database to be continually updated and improved as new information becomes available and for original data sources to be identified when conflicting or erroneous information about a well is uncovered or if further information from the original data source is desired. Although it is still in development, the geothermal well database will eventually be used to help estimate in-field EGS resource potential in the U.S. Results and summary information from the master geothermal well database is currently expected to be publically available at the end of 2014

    Chemotherapy and Stem Cell Transplantation Increase p16

    AbstractThe expression of markers of cellular senescence increases exponentially in multiple tissues with aging. Age-related physiological changes may contribute to adverse outcomes in cancer survivors. To investigate the impact of high dose chemotherapy and stem cell transplantation on senescence markers in vivo, we collected blood and clinical data from a cohort of 63 patients undergoing hematopoietic cell transplantation. The expression of p16INK4a, a well-established senescence marker, was determined in T-cells before and 6months after transplant. RNA sequencing was performed on paired samples from 8 patients pre- and post-cancer therapy. In patients undergoing allogeneic transplant, higher pre-transplant p16INK4a expression was associated with a greater number of prior cycles of chemotherapy received (p=0.003), prior autologous transplantation (p=0.01) and prior exposure to alkylating agents (p=0.01). Transplantation was associated with a marked increase in p16INK4a expression 6months following transplantation. Patients receiving autologous transplant experienced a larger increase in p16INK4a expression (3.1-fold increase, p=0.002) than allogeneic transplant recipients (1.9-fold increase, p=0.0004). RNA sequencing of T-cells pre- and post- autologous transplant or cytotoxic chemotherapy demonstrated increased expression of transcripts associated with cellular senescence and physiological aging. Cytotoxic chemotherapy, especially alkylating agents, and stem cell transplantation strongly accelerate expression of a biomarker of molecular aging in T-cells

    Flooding Events Post Hurricane Harvey: Potential Liability for Dam and Reservoir Operators and Recommendations Moving Forward

    When Hurricane Harvey hit the Texas coast as a category 4 hurricane on August 25, 2017, it resulted in $125 billion in damage, rivaling only Hurricane Katrina in the amount of damage caused. It also resulted in the deaths of 88 people and destroyed or damaged 135,000 homes. Much of that devastation was the result of flooding. The storm dumped over 27 trillion gallons of rain over Texas in a matter of days. Some parts of Houston received over 50 inches of rainfall. The potential liability that dam and reservoir operators may face for decisions they make during storm and flooding events has now become a major concern for Texas citizens and its elected officials. Law suits have now been instituted against the federal government for its operation of two flood control reservoirs, as well as against the San Jacinto River Authority for its operation of a water supply reservoir. Moreover, the issues and concerns have been placed on the agenda of a number of committees preparing for the 2019 Texas legislative session. This report reviews current dam and reservoir operations in Texas and examines the potential liability that such operators may face for actions and decisions taken in response to storm and flooding events. In Section III, the report reviews dam gate operations and differentiates between water supply reservoirs and flood control reservoirs. It also considers pre-release options and explains why such actions are disfavored and not recommended. In Section IV, the report evaluates liabilities and defenses applicable to dam and reservoir operators. It explains how governmental immunity can limit the exposure of state and federally-run facilities to claims seeking monetary damages. It also discusses how such entities could be subject to claims of inverse condemnation, which generally are not subject to governmental immunity, under Texas law as well as under the Fifth Amendment to the U.S. Constitution. In addition, the Section discusses negligence and nuisance claims and concludes that plaintiffs asserting either or both of these claims will have difficulty presenting successful arguments for flooding-related damage and harm against operators who act reasonably in the face of storm-related precipitation. Finally, Section V offers recommendations that dam and reservoir operators might pursue in order to engage and educate the public and thereby reduce the potential for disputes and litigation. Specifically, the report highlights the need for expanded community outreach efforts to engage with municipalities, private land owners, and the business community in flood-prone neighborhoods both below and above a dam. It also recommends implementation of proactive flood notification procedures as a way of reaching and alerting as many people as possible of potential and imminent flooding events. Finally, the report proposes implementation of a dispute prevention and minimization mechanism and offers recommendations for the design and execution of such a program

    The Hybrid Airship Multi-Role (HAMR) Anti-Submarine Warfare (ASW) Mission Module project applies established systems engineering principles and processes to the design of an ASW payload module that examines the capability of the HAMR to perform persistent ASW mission support. Critical system functions and objectives are identified and are assigned appropriate quantitative metrics. Additionally, three alternative architectures are generated and evaluated using the appropriate metrics based on results from modeling using Naval Systems Simulation (NSS). Manning is considered as a key stakeholder parameter and is included as an evaluation concern. The alternatives are also compared through the examination of life cycle costs.     International consensus on (ICON) anaphylaxis

    ICON: Anaphylaxis provides a unique perspective on the principal evidence-based anaphylaxis guidelines developed and published independently from 2010 through 2014 by four allergy/immunology organizations. These guidelines concur with regard to the clinical features that indicate a likely diagnosis of anaphylaxis -- a life-threatening generalized or systemic allergic or hypersensitivity reaction. They also concur about prompt initial treatment with intramuscular injection of epinephrine (adrenaline) in the mid-outer thigh, positioning the patient supine (semi-reclining if dyspneic or vomiting), calling for help, and when indicated, providing supplemental oxygen, intravenous fluid resuscitation and cardiopulmonary resuscitation, along with concomitant monitoring of vital signs and oxygenation. Additionally, they concur that H1-antihistamines, H2-antihistamines, and glucocorticoids are not initial medications of choice. For self-management of patients at risk of anaphylaxis in community settings, they recommend carrying epinephrine auto-injectors and personalized emergency action plans, as well as follow-up with a physician (ideally an allergy/immunology specialist) to help prevent anaphylaxis recurrences. ICON: Anaphylaxis describes unmet needs in anaphylaxis, noting that although epinephrine in 1 mg/mL ampules is available worldwide, other essentials, including supplemental oxygen, intravenous fluid resuscitation, and epinephrine auto-injectors are not universally available. ICON: Anaphylaxis proposes a comprehensive international research agenda that calls for additional prospective studies of anaphylaxis epidemiology, patient risk factors and co-factors, triggers, clinical criteria for diagnosis, randomized controlled trials of therapeutic interventions, and measures to prevent anaphylaxis recurrences. It also calls for facilitation of global collaborations in anaphylaxis research. In addition to confirming the alignment of major anaphylaxis guidelines, ICON: Anaphylaxis adds value by including summary tables and citing 130 key references.
